siRNA(「エスアイアールエヌエー」と読みます；small interfering RNAの略です)を用いてRNAi(RNA interference)を起こして目的のmRNAをノックダウンすることが哺乳類に対してもできることが最近わかってきました。 siDirect（「エスアイダイレクト」と読みます）はそのsiRNAをデザインする日本発のツールです。インターフェースや説明書きがすべて英語となっており日本語化されていないので素人にはなじみにくいかもしれませんが、設計したsiRNAが他の遺伝子に影響を及ぼさないか他の遺伝子配列にたいする配列類似性検索も同時に行うことができ、便利なツールとなっています。 前回の動画ではsiRNAの設計の基本を紹介しました。siDirectはいくつかの設計オプションを設定することができます。また、設計オプションを使用することでshRNAの設計をすることもできます。今回の動画では、siDirectのオプションの使い方およびshRNAの設計法について紹介します。
